Abstract
: A sensitive, specific, accurate and precise phenol鄄sulfuric acid method was established for
quantitative assay of polysaccharides in
Zanthoxylum bungeanum
leaves. And polysaccharides in leaves of
Dahongpao, a cultivar of
Z. bungeanum
in Shaanxi province, were successfully determinated by the pro鄄
posed phenol鄄sulfuric acid method. Galactose was used as standard monosaccharide and 484nm was cho鄄
sen as working wavelength. The optimum conditions for chromogenic reaction were obtained by single fac鄄
tor experiments and orthogonal test: addition sequence, phenol鄄sample鄄concentrated sulfuric acid; tem鄄
perature of chromogenic reaction, 25 益; amount of 5% phenol, 0郾 3 mL; amount of concentrated sulfuric
acid, 3郾 5 mL; and duration of chromogenic reaction, 30 min. System suitability test indicated that re鄄
gression equation of standard curve was

accuracy and precision was 10郾 00 - 60郾 00 滋g / mL. LOD and LOQ were 2郾 08 滋g / mL and 6郾 30 滋g / mL,
respectively. Intra鄄and inter鄄day precision varied from 0郾 49% to 3郾 64% and from 5郾 17% to 6郾 05% ,
respectively. And mean recovery was 101郾 19% . The colored sample solution was stable within 2 h. The
content of polysaccharides in Dahongpao leaves in Shaanxi province, was in the range of 8郾 11 - 8郾 89 mg / g.
The developed colorimetric method provides a basis for quantification of polysaccharides from
Z. bungea鄄
num
leaves and exploitation of leaf resources.
