近年来,我国经济快速发展,人民生活水平和预期寿命显著提高,然而人口老龄化加剧、居民生活节奏加快、不健康的饮食和生活方式盛行等社会现象也伴随而来,导致居民慢性疾病等健康风险增加,国民大健康形势日益严峻。当前,心脑血管疾病、神经退行性疾病、抑郁症等疾病的发病率也逐年升高,严重影响了居民的生活品质以及社会的正常运行。“健康中国2030”国家战略对食品产业提出了新的要求,强调通过营养干预的方式降低慢性病的发病率,提升国民健康水平。
功能性油脂由于其脂肪酸组成、结构的特点,不仅能够满足人们的基本营养需求,还能预防疾病和促进健康,近年来成为食品营养学领域的研究热点。ω-3多不饱和脂肪酸(omega-3 polyunsa-turated fatty acids, ω-3 PUFAs)是重要的功能性高值脂肪酸,具有促进神经系统生长发育、抗炎和抗氧化、提高免疫力、抗血栓形成、降低血脂等功效,且对神经系统疾病和情绪障碍等具有显著改善作用,现已广泛应用于特殊膳食、健康食品、化妆品及药品等产业。本文拟结合近年来ω-3 PUFAs改善人体健康的最新研究,对ω-3 PUFAs在我国食品领域的发展方向和需要突破的技术难点进行梳理,并提出建议,以期为功能性油脂产业的发展提供理论参考。
按脂肪酸碳链长度及所含双键的数量和位置进行分类,含有一个以上双键的脂肪酸称为多不饱和脂肪酸,根据第一个双键的甲基位置进一步可将PUFAs分为ω-3、ω-6、ω-7、ω-9等系列[1]。ω-3 PUFAs是机体内参与多种代谢过程的生物必需大分子物质,包括α-亚麻酸(α-linolenic acid, ALA)、二十碳五烯酸(eicosapentaenoic acid, EPA)、二十二碳六烯酸(docosahexaenoic acid, DHA)等。ω-3 PUFAs属长链脂肪酸,摄入后消化过程较为复杂。PUFAs的消化吸收、转运过程见图1。首先,含有ω-3 PUFAs的脂质食物进入口腔,在咀嚼和舌脂肪酶参与的条件下被初步分解,通过食道被推进到胃,并在胃液中被胃脂肪酶进一步水解,胃中预消化的脂质被推入十二指肠,被胰脂肪酶水解和消化。最终,脂类的消化产物,包括单甘油酯、胆固醇、游离脂肪酸等,进一步被胆汁盐和磷脂乳化,从而促进其在体内的吸收和转运[2]。在整个消化过程中,脂质消化率随脂肪酸链长度的增加而降低,消化率由高到低依次为短链脂肪酸、中链脂肪酸和长链脂肪酸[3]。ω-3 PUFAs以甘油三酯的形式通过乳糜微粒转运到肝脏,一些ω-3 PUFAs会以血浆磷脂的形式从肝脏释放到体液循环,进入全身的细胞膜磷脂中;另一些ω-3 PUFAs以甘油三酯的形式储存在脂肪组织中[4]。尽管ω-3 PUFAs对生命健康至关重要,但人体缺乏Δ-9去饱和酶,不能内源性合成ALA,只能通过食物摄取,也只有少量的ALA可以在体内转化为EPA或DHA,在机体内作为一些内源性分子的前体。EPA和DHA的体内生物合成效率很低,成年男性的EPA转化率大约为7%,DHA的转化率只有0.01%,远不能满足营养和生理需求,因此人体主要通过摄入海洋多脂鱼类或膳食补充剂来获得这类必需脂肪酸,也称为人类日常必须摄入脂肪酸。

1.2.1 动植物来源
膳食中的ω-3 PUFAs主要来源于海洋鱼类和富含油脂的植物,如坚果、种子[5],见表1。大多数作物种子和植物油,例如菜籽油、大豆油、玉米油和葵花籽油等,存在大量亚麻酸(linolenic acid,LA)形式的ω-6 PUFAs,存在少量的ALA。一些绿叶蔬菜中,整体油脂含量较低,但总脂肪酸的60%~70%为ALA。水产、畜肉能够提供EPA以及DHA。在植物和动物性食品中,约98%的长链PUFAs以三酰基甘油(triacylglycerol, TAGs)的形式存在,其次是磷脂(phospholipids, PLs)和二酰基甘油(diacylglycerol, DAGs),以及少量的胆固醇酯和脂溶性维生素酯。不同形式的ω-3 PUFAs在人体中表现出显著不同的生物利用度[6]。与TAGs形式相比,由于PLs具有两亲性、高水分散性和对磷脂酶更高的敏感性,因而具有更高的生物利用度。此外,由于大脑对PLs的摄取更高,因此PLs形式的ω-3 PUFAs补充剂比TAGs更为有效。例如,磷虾油中DHA近35%以PLs的形式存在,因此其被认为比鱼油更有效[7]。从海洋生物中提取ω-3 PUFAs成本高,且易产生资源浪费和环境污染,受季节、地理位置等外部因素影响大。以现在的深海渔业资源为例,世界主要商业鱼类已处于过度捕捞状态,且海洋污染严重,导致渔业资源的日益枯竭,鱼油价格飞涨。

1.2.2 微生物来源
许多微生物具有脂肪酸相关的代谢、转化途径,具有生产ω-3 PUFAs的潜力,见图2。

1)微藻。微藻是一种能够进行光合作用的绿色细胞工厂,不仅会显著影响CO2的固定,且其生产过程全程可控,是可持续生产高价值化合物的平台。Rodolfi等[8]的研究表明,微藻可以捕获1.8倍自身质量的CO2,能够显著抑制温室气体的排放。微藻的生长繁殖速度快,培养周期短,且具有强大的脂质储存能力,是生产多不饱和脂肪酸的潜力菌株。研究显示,微藻中含有的脂质通常占其干重的1%~40%,目前已经被开发为多不饱和脂肪酸的新来源,包括LA、ARA、EPA和DHA[9]。《中华人民共和国食品安全法》和《新食品原料安全性审查管理办法》规定,莱茵衣藻、螺旋藻、蛋白核小球藻、盐藻、雨生红球藻、拟微球藻、裂壶藻等多种微藻的DHA藻油已经获得新资源食品的认证。此外,与海洋动物来源相比,微藻生产的多不饱和脂肪酸更易于被人体吸收和代谢,具有较高的生物利用度和稳定性,且其生产过程全程可溯源,这也确保了微藻用于生产多不饱和脂肪酸的安全性和可靠性。
微藻中多不饱和脂肪酸的合成通常可以通过改变发酵条件和基因工程手段进行控制。在发酵调控方面,微藻的脂质代谢可以通过调节生长参数来控制,如温度、光照、CO2和培养基成分,这些参数的改变可以提高生长速度,从而提高微藻的生物量和脂质生产能力。其中,温度的改变在微藻调节其细胞膜的流动性方面起重要作用。通常,较低的温度会增强脂肪酸的不饱和度,Almeydo等[10]在Cylindrotheca closterium的生长阶段将温度降低,刺激该菌株产生了更高含量的多不饱和脂肪酸。然而,并不是所有微藻都遵循这样的规律,Converti等[11]将温度从20 ℃提高到25 ℃,Nannochloropsis oculata的脂质含量反而增加[11]。此外,培养基的组成也会影响微藻的脂质代谢,其中用于提高微藻中脂质含量的常见策略包括限氮、限磷等[12]。氮源是影响微藻生长和脂质积累的关键营养物质之一,许多研究证明,限氮条件可用于增加Chlorella emersonii、Neochloris oleoabundans和Chlorella sorokiniana等微藻物种的TAG积累[13-15]。与限氮条件相似,限磷也可以促进微藻中脂质含量和TAG含量的增加,如Scenedesmus sp.[16]、Isochrysis galbana[17]和Pavlova lutheri[18]。基因工程技术的进步给微藻生产多不饱和脂肪酸提供了另一种可能,由于缺少通用基因编辑工具,大多数研究都集中在模式微藻菌株的遗传改造上,如灰藻(Phaeodactylum tricornutum)、拟微球藻(Nannochloropsis oceanica)和莱茵衣藻(Chlamydomonas reinhardtii)。Peng等[19]在灰藻中过表达Δ5去饱和酶后,多不饱和脂肪酸含量增加,其中,EPA含量提高了58%;Poliner等[20]通过在微拟球藻中过表达Δ12去饱和酶和Δ5去饱和酶后,也能够促使其EPA含量提高。代谢工程的应用加速了微藻中多不饱和脂肪酸含量的积累。Hamilton等[21]在灰藻中异源引入绿藻(Ostreococcus tauri)来源的Δ6去饱和酶和Δ5延长酶,显著增加了DHA含量。无论是发酵调控手段还是基因工程手段,都显示了微藻用于生产多不饱和脂肪酸的巨大前景。
2)丝状真菌。丝状真菌为单细胞真菌,由菌丝和孢子两部分组成。丝状真菌作为一种重要的工业微生物,能够利用多种廉价碳源、氮源生物转化为多种高附加值化学品[22]。很多丝状真菌,如高山被孢霉(Moriterella alpina)、华丽腐霉(Pythium splendens)、长被孢霉(Mortierella elongata)、轮枝霉(Diasporangium)、畸雌腐霉(Pythium irregulare)等,都可以天然生物合成多种PUFAs。其中,被孢霉属的成员是产油能力较强的真菌,胞内存在多种延长酶/去饱和酶,在PUFAs生产方面具有很大优势,可以天然生产的脂肪酸有GLA、EPA、ARA等[23]。Bajpai等[24]发现,高山被孢霉在25 ℃和11 ℃的低温条件下能够合成EPA,且EPA的产量随着亚麻籽油浓度的增加而增加。除了高山被孢霉菌,Linnemannia hyalina也具有潜在的脂质生产能力,天然可生产高水平的油酸[25]。为了提高华丽腐霉中EPA的含量,Ren等[26]在华丽腐霉RBB-5菌株中过表达了EPA合成的限速酶,即Δ6去饱和酶、Δ12去饱和酶和Δ17去饱和酶,EPA产量达到1.43 g/L,是对照菌株的10倍。这些研究表明了丝状真菌在生产PUFAs方面的巨大潜力。
3)产油酵母。脂质积累超过其细胞干重的20%的酵母通常被定义为产油酵母[27],例如,圆红冬孢酵母(Rhodotorula toruloides)、解脂耶氏酵母(Yarrowia lipolytica)、担子菌酵母(Rhodosporidiobolus fluvialis)等。与其他产油微生物相比,产油酵母具有生长周期短,可利用底物广泛,基因操作工具丰富等优势[28]。解脂耶氏酵母是最具代表性的产油酵母,是被美国食品药品监督管理局认证的“公认安全”菌株,可以天然积累大量的脂质(占细胞干重的30%)。野生型解脂耶氏酵母只能产十六碳酸和十八碳酸,因此必须通过代谢工程改造才能在解脂耶氏酵母中从头合成其他的长链脂肪酸[29]。Imatoukene等[30]通过在解脂耶氏酵母JMY3479过表达高山被孢霉来源的Δ12去饱和酶并敲除β-氧化途径,提高了亚油酸的产量。Gemperlein等[31]将人工PUFA生物合成基因簇PKS途径引入解脂耶氏酵母Po1h中,成功实现DHA的异源合成。Xie等[32]在解脂耶氏酵母Z7344菌株中异源表达EPA合成途径基因,并结合两阶段连续发酵工艺策略,增加了EPA占总脂肪酸的比例。在人工智能的指导下,理性计算EPA途径基因的最佳拷贝数组合,在短时间内构建了EPA含量高达59%的解脂耶氏酵母工程菌株[33]。解脂耶氏酵母也是第一个有潜力实现微生物生产EPA作为鱼油替代品的商业化宿主菌株。除了解脂耶氏酵母,圆红冬孢酵母也是生产油脂的优秀宿主, 在营养限制条件(氮、磷酸盐和硫酸盐)下,圆红冬孢酵母积累的脂质产量高达细胞干重的70%[34]。许多研究人员通过诱变、发酵优化和代谢工程等策略提高圆红冬孢酵母油脂积累能力[35-36]。油酸是圆红冬孢酵母中最丰富的脂肪酸之一,过表达Δ9去饱和酶促进硬脂酸向OA的转化,提高了OA的产量,这也促进了其在生物柴油、生物润滑剂等方面应用[37]。此外,通过外源引入Δ12去饱和酶和Δ6去饱和酶基因可成功生产GLA[38]。但是圆红冬孢酵母生产其他PUFAs的研究较少,因此,需要进一步揭示胞内脂肪酸生产和生物过程机制,开发丰富的遗传工具,从而实现多种PUFAs的定制化生产。
越来越多的研究证明,ω-3 PUFAs对健康有益,尤其是EPA和DHA,ω-3 PUFAs的健康功效及机制见图3。ω-3 PUFAs在降低心血管疾病(cardiovascular disease, CVD)风险上最早被认可,包括调节甘油三酯水平、缓解高血压、改善炎症,以及改善心脏和血管功能等[39]。此外,ω-3 PUFAs在促进婴儿、儿童和青少年的视觉和脑功能发育方面发挥着重要作用[40-41]。由于EPA和DHA是抗炎介质前体,有助于缓解神经退行性疾病和精神障碍的症状,如阿尔茨海默病、帕金森病和抑郁症[42]。ω-3 PUFAs的其他健康益处包括降低感染风险,预防自身免疫性疾病,治疗严重创伤,缓解氧化应激与细胞凋亡,调节血小板活性,降低糖尿病、癌症风险等[43-49]。

20世纪70年代,研究者发现格陵兰因纽特人的心血管疾病发病率较低,与食用大量的海洋鱼类等海洋生物有关。后续研究得出了鱼类摄入量或膳食中补充ω-3 PUFAs与心脏病和中风等心血管相关疾病的发生率之间存在显著关联[50]。摄入含有ω-3 PUFAs的鱼油能够通过降低血压和甘油三酯水平、抑制血小板聚集以及其他许多有益作用来降低心血管疾病风险以及心血管疾病死亡率[51-52]。值得注意的是,一项研究指出,EPA单独补充减少了25%的心血管相关疾病的发生率,包括死亡、心肌梗死、中风和心绞痛等[53],并且EPA单独使用比EPA和DHA联合治疗具有更好的效果[54]。EPA被誉为“血管清道夫”,利用EPA进行炎症干预实验中,发现体内炎症关键指标脂蛋白a显著降低,因此EPA对高血压、脑血栓、动脉粥样硬化等心脑血管疾病的发生有重要的防治作用[55-56]。此外,EPA乙酯剂(商品名Epadel和Vascepa)已被美国食品药品监督管理局批准作为阻塞性动脉硬化和高甘油三酯的治疗剂[57]。最近的一项小型随机临床研究表明,在CVD的某些指征上,如炎症方面:在减轻慢性炎症上,DHA比EPA更有效;在平衡促炎和抗炎因子方面,EPA更有效[58]。因此,EPA和DHA的有益作用似乎是不同的,这一发现具有重要的临床应用价值,需要进一步的研究来验证EPA和DHA在调节炎症反应和心血管健康方面的不同作用。
大量流行病学研究表明,长期摄入地中海饮食(富含海鲜、水果、蔬菜和橄榄油等)与老年人群的认知功能呈正相关[59-62]。在几项纵向研究中,发现食用鱼类来源的ω-3 PUFAs摄入量的增加与认知能力下降、阿尔茨海默病(Alzheimer’s disease, AD)和痴呆的发病风险降低有关[63-64]。膳食补充ω-3 PUFAs已成为一种常见的维持认知功能的补充和替代医学治疗的方式。2010年,美国国立卫生研究院专家共识意见指出,营养和饮食是影响神经退行性疾病风险的重要因素[65]。为了阐明ω-3 PUFAs在改善认知功能并最终预防或延缓AD发病方面的作用,研究人员进行了多项临床试验。大脑生理和病理变化可能在AD确诊前10~20年就已经开始,生理和病理变化一旦开始,就很难停止、减缓或逆转。在目前的临床研究中,DHA主要对健康老年人和轻度认知障碍患者具有潜在的保护作用,而支持其对AD患者有益的证据仍然有限。在动物和细胞研究中,DHA发挥认知益处的各种机制包括,抗淀粉样蛋白Aβ活性、降低Tau蛋白磷酸化、抗炎和抗氧化,以及其作为核受体配体的作用,进而影响下游基因表达。DHA补充对携带载脂蛋白E4(apolipoprotein E, ApoE4)基因的健康老年人会产生有益影响,可能由于ApoE4携带者脑中摄取和利用DHA的能力下降[66-67]。这些差异的潜在机制可能涉及ApoE4携带者循环DHA分解代谢增加,血脑屏障功能受损,阻碍DHA运输到大脑。另外,体育锻炼和认知刺激结合ω-3 PUFAs补充可以有效减缓体弱老年人的认知能力下降[68-69]。在另一项研究中,ω-3 PUFAs与番茄红素、鼠尾草酸或叶黄素联合使用,发挥了协同抑制神经炎症的作用[70]。ω-3 PUFAs与类胡萝卜素和维生素E联合使用,改善了轻度认知损伤患者的情景记忆和整体认知[71]。
研究表明,DHA占人脑总脂肪酸的10%~15%,是中枢神经系统中膜的结构成分,可以通过调节信号转导途径,影响神经传递、细胞突触的延伸和再生来影响神经功能,而DHA在大脑中的积累发生在孕晚期到2岁,后续其内源性形成则相对较低[17]。Hoffman等[72]证明,喂食补充DHA配方奶粉的早产儿和足月儿比未补充DHA的对照组的视力和神经发育会更好。儿童摄入足够的DHA则可以增强智力、注意力和记忆力,降低罹患系统性疾病的风险。研究显示,每天摄入1 183 mg DHA的儿童智商会更高,每天使用500 mg较低剂量的DHA可以增强神经发育[73-74]。1996年,美国婴幼儿配方奶粉中开始添加纯植物来源的DHA,2000年,我国首款添加DHA奶粉上市;为了规范DHA的使用,1999年,联合国粮食及农业组织、世界卫生组织率先提出婴儿配方奶粉中必须含有DHA,并给出了孕期及哺乳期妇女和幼儿的推荐摄入量;2010年,世界卫生组织再次颁布国际标准,规定婴幼儿配方奶粉中DHA含量不达标视为不合格;2011年,美国医学研究所发布了各类人群DHA每日适宜摄入量,再次拓宽了DHA的适用范围;随后,澳大利亚国家健康医学研究委员会、欧盟等国际组织针对DHA在预防心血管疾病、慢性疾病等方面也做出了相应的规定;2021年,我国国家卫生健康委员会再次提出1段、2段婴儿配方食品中DHA含量至少需要达到3.6 mg/100 kJ,使婴幼儿配方奶粉中DHA的含量标准再次提高。因此,在婴儿配方奶粉中添加ω-3 PUFAs,尤其是DHA,已成为一种全球惯例。
鱼类的食用与重度抑郁症的发生存在显著相关性[75]。在一些临床研究中发现,9.6 g/d的EPA与DHA混合摄入可改善抑郁症[76]。值得注意的是,更高剂量EPA的补充与抑郁症症状的改善有关[77]。有研究表明,ω-3 PUFAs 的摄入量与乳腺癌发生率、结直肠癌发生率呈负相关[78-79]。ω-3 PUFAs对癌症预防和治疗的作用是复杂的,有证据表明,在肿瘤细胞内,环氧酶2(COX-2)的活性显著增强,会促进PGE2的合成从而诱导细胞增殖并刺激BCL-2蛋白的表达。BCL-2会造成细胞凋亡以及生长失衡,从而促进肿瘤的发生,而EPA能够抑制COX-2的合成从而在一定程度上抑制肿瘤的发生[80]。然而,还需要进一步的研究和更加精确设计的临床试验来证明ω-3 PUFAs对各种癌症风险的影响。ω-3 PUFAs在孕妇怀孕期间起着重要作用,能够确保正常怀孕的进展。研究者通过调查孕妇怀孕期间是否补充EPA和DHA发现,儿童出生时EPA和DHA水平越高,神经发育越好,这也提示了ω-3 PUFAs能够对神经发育产生有利影响。另外,ω-3 PUFAs在创面愈合和组织修复过程中起着重要作用,可抑制炎症反应、促进血管生成、调节神经系统、控制血糖水平、抑制微生物生长、加速创面愈合[81]。
药用级EPA和DHA在治疗高甘油三酯血症方面的成功,也增加了日常居民消费ω-3 PUFAs的动力。在食品中,ω-3 PUFAs的添加可以通过不同的途径实现:1)直接添加纯化的ω-3 PUFAs油;2)在食物中添加富含ω-3 PUFAs的食品配料如鱼粉等;3)通过给动物喂食富含ω-3 PUFAs的饲料,富集动物制品中的ω-3 PUFAs。
Schizochytrium sp.可以产生高产量的DHA或EPA。根据不同脂质组成情况,可以从其中分离提取应用于指定食品的ω-3 PUFAs藻油。婴儿配方奶粉是ω-3 PUFAs油最主要的用途,目前占据着 ω-3 PUFAs 藻油市场的最大份额。除婴儿配方奶粉外,ω-3 PUFAs藻油也应用于营养强化食品,如液态奶和酸奶、肉制品、香肠等。在这些产品中,ω-3 PUFAs藻油是以水包油乳液的形式使用,即使在低pH值环境下,也能在食品加工和储存过程中提供更好的抗氧化稳定性[82]。
由于存在多个双键,ω-3 PUFAs稳定性较差,氧化降解会产生自由基和氧化产物,产生异味并且降低营养属性。因此,通常需要在精制 ω-3 PUFAs 油中添加抗氧化剂,以提高氧化稳定性,延长保质期。与传统的合成抗氧化剂相比,天然抗氧化剂具有明显的安全性优势。由于不同抗氧化剂的作用机制存在较大差异,例如可以通过清除自由基和淬灭单线态氧发挥作用,也可以通过去除金属离子发挥作用。通常在ω-3 PUFAs油的保存中使用抗氧化鸡尾酒法,复配抗坏血酸棕榈酸酯、维生素E、植物酸和茶多酚组合抗氧化,效果优于单独添加每种抗氧化剂[83]。其他天然抗氧化剂如卵磷脂、迷迭香提取物、槲皮素、绿原酸、咖啡酸也是在ω-3 PUFAs油的保存中经常使用的[84]。另外,利用微胶囊技术,将ω-3 PUFAs油包裹在外壳材料层中,也能够在食品加工和储存过程中减轻脂肪酸氧化。使用此技术还可以增加 ω-3 PUFAs 在体内的生物利用度。
在水产和动物饲料中添加ω-3 PUFAs,能够增强动物本身的免疫功能,提高生长速度,提升肉品质,其中,鱼类最容易富集ω-3 PUFAs。目前,在水产养殖中添加EPA和DHA已经非常常见,但是,这种富集能力是物种特异性的,与动物的脂质水平有关。另外,通过ω-3 PUFAs藻粉饲喂奶牛能够显著提高牛奶中的ω-3 PUFAs,类似的,在鸡蛋中也可以按照此方式富集ω-3 PUFAs。在羔羊、山羊、猪和公猪饲料中添加ω-3 PUFAs藻粉均能够提高肉制品中ω-3 PUFAs的含量。消费者通过食用动物制品富集的ω-3 PUFAs,在安全性、生物利用度方面具有显著的优势。
ω-3 PUFAs作为一种重要的功能性脂肪酸,对人体健康的作用已得到了广泛的研究和认可。然而,关于ω-3 PUFAs的研究仍有许多方面值得深入探讨,未来ω-3 PUFAs的研究重点应在如下领域:1)更精准的分子靶点,深入研究ω-3 PUFAs在细胞水平上的作用机制,明确其与各种疾病相关的分子靶点,形成更有针对性的、功效更为突出的药物制剂或功能性食品;探讨ω-3 PUFAs与人类基因表型之间的关系,为不同人群的膳食补充提供更深刻的依据;研究肠道菌群在ω-3 PUFAs代谢和生物利用度中的作用,揭示肠道微生物与 ω-3 PUFAs 健康效应之间的关系。2)结合合成生物学、人工智能、计算生物学等多个学科,挖掘不同底盘菌株作为多不饱和脂肪酸合成的新资源,获得简便高效的遗传操作工具,深度解析多不饱和脂肪酸合成机制,开发食物中多不饱和脂肪酸定制化平台,以满足不同年龄层次对不同多不饱和脂肪酸的需求,开发新构型、高纯度的 ω-3 PUFAs 产品。3)开发适用于不同人群的 ω-3 PUFAs 的功能食品形式及完善相应的活性保持技术。
