双水相萃取韭籽粕多糖的工艺及其性质研究

    摘要:

    为研究双水相萃取法对韭籽粕中提取多糖的效果,本文采用聚乙二醇/硫酸铵双水相系提取韭籽粕多糖并初步探讨韭籽粕多糖的抗氧化活性。通过单因素实验和Design-Expert8.06进行响应面设计对萃取技术工艺条件进行优化,得出最佳工艺参数为:PEG分子质量为6000,PEG质量分数为14.50%,硫酸铵质量分数为20.46%,在此条件下多糖的萃取率可达85.39%。在最佳工艺条件下,测定韭籽粕多糖对DPPH.、.OH的清除能力以及总还原力,结果表明韭籽粕多糖具有抗氧化活性,随着韭籽粕多糖浓度的提高,抗氧化活性增强。

    Abstract:

    The optimum extraction conditions of polysaccharides from Chinese leek seed meal by using Polyethylene glycol/ammonium sulfate diaqueous system were investigated using the Response Surface Method (RSM) and the antioxidant activity of extract was obtained under optimum extraction conditions. The results indicated that the optimum extraction conditions were PEG molecular mass was 6000, PEG mass fraction was 14.50%, ammonium sulfate mass fraction was 20.46%. Under the optimal conditions, the extraction rate of polysaccharides from Chinese leek seed meal was 85.39%. Antioxidant activity of the polysaccharides from Chinese leek seed meal, including the DPPH., .OH and reducing power measure. The results showed that the leek polysaccharides possessed antioxidant properties in vitro. And with the concentration of polysaccharides increasing, its antioxidant activity gradually becomes stronger.
